Support Response Form: I am the defendant on a summons of "Petition For Support and Other Relief". I need to file a response with the court, and would like to know if there is a specific form I use.
We do not have an existing support order, this will be the first. All I can find are forms for modifying support.

The form that you need to file is Florida Supreme Court approved Form 12.903(e) Answer to Supplemental Petition; which can be found on the Supreme Court's website at www.flcourts.org.
Instructions for that form state in part: This form should be used when you are responding to a supplemental petition for modification of Parenting Plan, time-sharing schedule, child support, or alimony.
This form is used to admit or deny all of the allegations in the supplemental petition if you do not plan to file a counterpetition. There is no form for a counterpetition to a supplemental petition in these Family Law Forms.
I realize you stated in your question that you need to respond to an original petition for support and other relief -- and not a modification.
The title of this form makes it appear that it is only suitable for responding to a petition for modification.
However, in reading the form, you can see that it will suit your purpose.
